GelRed is a sensitive, stable and environmentally safer alternative to the highly toxic ethidium bromide (EB) for staining dsDNA, ssDNA or RNA in agarose or polyacrylamide gels (see PAGE GelRed Nucleic Acid Gel Stain). The GV3101 strain has a C58 chromosomal background with rifampicin resistance and the Ti plasmid pMP90 (pTiC58DT-DNA) with gentamicin resistance. The GV3101 Ti plasmid has the T-DNA region sequences deleted and transformation with a binary vector containing the missing T-region results in a functional T-DNA binary system that allows for transfer of genetic material into a host plant’s genome. Gel Loading Dye, Orange (6X) is a pre-mixed loading buffer with a tracking dye for agarose and non-denaturing polyacrylamide gel electrophoresis. This solution contains SDS, which often results in sharper bands, as some restriction enzymes are known to remain bound to their DNA substrates following cleavage. EDTA is also included to chelate magnesium (up to 10 mM) in enzymatic reactions, thereby stopping the reaction. Reconstitute each pack with 900 ml deionized water, then add 100mL of either methanol or ethanol to make 1X transfer buffer. Do not use acid or base to adjust the pH.
